November Sales Disappoint; What Happened to Black Friday's 13 ...

?Sales at stores open at least a year declined in November at major American store chains, including Macy?s, Nordstrom, Kohl?s and Target, sending a shiver through the retail world Thursday.

The reporting period included Thanksgiving and Black Friday, the official kickoff of the critical holiday shopping season. Early reports regarding those days had been mixed, and the individual retailers? dim results suggest a big challenge in the coming weeks for retailers.?

?

?

Thomson Reuters tracks major retailers actual sales results ? not surveys ? and the most recent data was disappointing. Sales at stores open at least a year gained 1.6% increase in November. This is far below the nonsensical 13% number the?National Retail Federation (NRF) trumpets to gullible journalists. It was even below the 3.3% increase consensus of analysts.

The NYT rounds up some of the sales figures, and they are not pretty:

Kohl?s sales -5.6% (vs expectations of +1.9%)

Target -1% (vs +2.1%)

Nordstrom -1.1% (vs +4.3%)

Macy?s -0.7% (vs + 1.5%)

There were a few winners:

Costco +6%

Limited? +5%

Gap +3%

We won?t get the final holiday sales figures til January, but you can be pretty comfortable with the premise that there will not be an increase of 13% of retail sales this holiday season.

The lesson here is pretty stark. Be very aware of what you accept as a data source. Understand the differences between hard numbers ? i.e., sales receipts ? and squishy emotional guesses produced by surveying consumers.

The bottom line:? The NRF is an industry shill group that produces nonsensical spin that misleads investors, fools the public, and bamboozles incompetent journalists. Any writer who uncritically reprints their nonsense deserves to be fired; any media outlets that publishes their junk should be put quarantined and put on your DO NOT READ list.

(AP) ? Court records show Mayim Bialik filed for divorce from her husband of nine years on the same day she announced the couple's split in a blog post.

She cited irreconcilable differences with husband Michael Stone in the documents filed Nov. 21 in Los Angeles.

Bialik currently stars on the CBS comedy "The Big Bang Theory" and rose to fame as the star of the TV show "Blossom."

She has been a proponent of "attachment parenting" and the former couple have two sons together, ages 7 and 4. Bialik has said their parenting style was not a factor in the divorce and she is seeking joint custody of the children.

The 36-year-old wrote in her post last week that the divorce is "terribly sad, painful and incomprehensible" for children.

Responsibilities Of Your Personal Injury Attorney LA: What Are They ...

Like any other professionals, lawyers also have certain responsibilities on their shoulders and personal injury lawyers are no exception to this. As for the responsibilities of personal injury attorney LA, their prime target is to get their clients what they deserve, a fair compensation. That?s not all; they also have various other responsibilities regarding serving their clients in the best way possible.

The responsibilities of personal injury lawyers encompasses both professional and ethical rules as well as codes of conduct that have been set forth by the bar associations of the state that have issued the incense to the lawyers. As soon as any lawyer gets his or her license from the bar council of the state he or she intends to practice in, the lawyer is legally permitted to practice law in his or her respected field, file cases, provide legal solutions to the clients who are supposedly the victims of personal injury, argue cases in the court and last but not least draft any type of legal documents related to the case.

Personal injury lawyers are often referred as plaintiff?s lawyers and their responsibilities are not limited to just filing of the case. they are required to interview their clients in order to understand and evaluate the case and go through every single details associated with the case, identify each of the issues deeply rooted in their clients complaint and find out the strong points which may help them win the case, dig deep into the matter and find out substantial evidences crucial for the case and interview the witnesses to make a strong case.

However, the ultimate responsibility of any LA personal injury lawyer is to serve justice to the clients and help them obtain the compensation they deserve to recover from the damages done to them. This is achieved through advocacy, verbal arguments in the court room, plaintiff counseling and advice on the legal issues related to the case. Besides, every personal injury lawyer should be sensible and sensitive enough to understand the suffering and pain of his or her clients and must pay personal attention to each and every case.

They should also adhere to the guidelines and ethics made certain by the bar association and abide by every word written in the book of law. Although the guidelines may vary from state to state the basic code of conduct remains the same for every lawyer and they should always evaluate the cases they undertake and exercise competence to ensure their clients get what they deserve.

Syrians risk lives in battle to protect ancient sites

Zain Karam / Reuters

A damaged ceiling is pictured in Bab Antakya district of Aleppo, Syria, on October 2, 2012. Aleppo's Old City is one of several World Heritage Sites in Syria that are considered at risk.

By Alastair Jamieson, NBC News

Editor's note: This story includes a correction.

Updated at 9:15 a.m. ET:?Even as civil war tears the nation apart, it seems Syrians can agree about one thing: The need to protect the country?s antiquities and World Heritage Sites that represent thousands of years of human history.

Rebel fighters and ordinary citizens are risking their lives to document the damage being done to Syria?s ancient?treasures and museums, according to Western monitors.

Now Bashar Assad's regime has joined in. Maamoun Abdul-Karim, director general of antiquities and museums, has launched a campaign, called "MySyria,"?(in Arabic) asking communities to help protect the nation?s cultural heritage from the civil strife.

All six World Heritage Sites have now suffered damage as the conflict widens, according to Emma Cunliffe, a volunteer monitor for the non-profit Global Heritage Fund.

One of oldest cities
Destruction includes heavy looting of temples and tombs in the trade city of Palmyra and a devastating fire in the medieval souk in Aleppo, one of the oldest continuously inhabited cities in human history.

World heritage body UNESCO?has led the outpouring of international concern.?Aleppo dates back to the 10th century B.C. and the present city is deemed to have "Outstanding Universal Value," by UNESCO.

Reuters, file

Demonstrators protest against Syria's President Bashar Assad in the ancient city of Palmyra on November 18, 2011.

"Pictures and video evidence gathered by people on the ground shows the extent of the damage and prove that none of these sites are now safe from the conflict," said Cunliffe, a postgraduate student at Britain's Durham University.

'Emergency red list' targets Syria's looted treasures

Looting, which led to the theft of many of Iraq's national treasures during the conflict that deposed Saddam Hussein, is also a risk in Syria.

"Large gangs of men turned up at Iraqi sites, totally overwhelming the protection, and looted on a vast scale. If that starts to happen in Syria there will be problems because there's little that can be done about it,? Cunliffe said.

More Syria coverage from NBC News

She said each side in the conflict blamed the other for damage to ancient buildings, but it was not easy to verify the claims.

Cunliffe said many people in Syria made films showing the damage being done to ancient sites.

She said that one man ?who uploaded most of the videos of the damage to the citadel of Qal'at al-Madiq in January to April stopped uploading when the government took the citadel/village in April. I have assumed the worst.?

New 'intelligence' body set to fight illicit trade in world's priceless treasures

Abdul-Karim hopes to encourage Syrians to prevent the war from causing permanent damage.

?The war in Syria has hit ... all aspects of life, including antiquities considered the common heritage of all Syrians, regardless of their thoughts or political alliances, whether loyalists or opposition,? Abdul-Karim told news website Al Akhbar following the campaign's launch.

He said there was also evidence of antiquities being smuggled out of the country.

'A loss to human civilization'
Dan Thompson, director of global projects at the Global Heritage Fund, said that there was little that could be done until the fighting stopped.

A Cluster Bomb reportedly dropped by Syrian government warplanes has killed up to 10 children as they played in a village on the outskirts of Damascus. Warning: There are distressing images. ITV's Bill Neely reports.

?The continuing damage and destruction of World Heritage Sites and other national antiquities in Syria during the present conflict is not only a loss to human civilization, but also greatly reduces the socio-economic potential these sites offer to local communities and the country as a whole,? he said in a statement.

"At present, unfortunately, the most anyone can do is to closely monitor and publicize the devastation ? and plead for both sides to respect the country?s cultural heritage, as UNESCO has done."

Samsung's report on Chinese suppliers makes for grim reading, especially between the lines

Samsung's audit of its Chinese suppliers makes for grim reading, especially between the lines

On the face of it, Samsung's latest report on working conditions at the factories of its Chinese suppliers follows just the kind of careful, lawyer-scrutinized language we'd expect from a big multinational. It repeats the manufacturer's earlier insistence that no children have been employed, while at the same time admitting that there have been "several instances of inadequate practices at the facilities" concerning workers being made to do too much overtime, not being given proper contracts, and being fined if they turn up late or are absent -- issues which had already been revealed at one supplier, and which Samsung promises to fix by the end of 2014.

Burrow further into the document, however, and Samsung's list of promised "corrective actions" implies that its internal investigation has uncovered evidence of other serious problems. These include "physical and verbal abuse," sexual harassment, a lack of first-aid equipment and inadequate safety training. Some information is also conspicuous by its absence -- at least in the short report linked below -- including clarity on how widespread any of these failings were among the 109 companies (and 65,000 employees) that have now been audited. Did they crop up at just a handful of factories, or were they endemic across China? We have no idea, but given how much data Samsung has now dutifully amassed, it surely does. We've asked the company for more detail and will update if we hear back.

Top 6 Adventure Trips in Grenada

Visiting the island of Grenada in the Caribbean doesn't mean that you'll have to lie around on a beautiful beach with the crystal clear waters lapping on the shoreline whilst you sip on an ice cold rum punch. There are plenty of opportunities for adventure and it will also give you a chance to see some of the most beautiful and memorable places in Grenada.

Here are my top tips for enjoying some Caribbean adventure whilst on the Spice Isle;

1. Trek the Jungle

The Grand Etang Lake and Forest Reserve is a magnificent landscape of rain forest, exotic flora and fauna. You'll see everything from hummingbirds to Mona monkeys to hibiscus bushes. It's also a great chance to take some excellent photographs too.

2. Slide Down the Waterfall at Mount Carmel

The walk to the waterfall is really quite short, but once you get there you'll be able to slide down a natural slide into the clear cool waters. Walk further along and you'll see a wall of water gushing down, perfect for a photographic backdrop.

3. Take a Fisherman's Boat Over to Sandy Island

This is regularly rated as one of the best adventures on the island. It's in the north and close to the town of Sauteurs. You'll be able to book this trip through one of the local hotels in Sauteurs. A great snorkel and you'll also be able to enjoy the freshly caught fish straight off the open fire and possibly drink from a fresh coconut.

4. Join the Hash House Harriers

These guys always go off the beaten path and pick routes that are suitable for both walkers and runners. You'll have to join in with the drinking at the end to really get the best of the event but you'll meet a wide range of people who live on the island and its well worth doing it.

5. Join in The Annual Carnival

There's nothing so much fun as a Caribbean Carnival. The highlight for many is J'Ouvert Morning, which is the opening of the event. If you know a local, all the better as they'll be able to show you the ropes on how to dance and enjoy the festivities like a local.

6. Watch the Leatherback Turtle Nest

Most visitors are absolutely astounded when they see these humungous marine creatures land on the beach. At over 1,000 lbs and sometimes over 6 foot long, they'll take your breath away as you watch them walk up the beach to lay their eggs.

Grenada and the Caribbean in general has a lot of activities to do that will give you some adventure and will allow you to see far more than you would on a traditional resort style holiday.

April Jones writes for Petite Anse Hotel in Grenada, West Indies. This small boutique hotel overlooks the Caribbean Sea all the way to the Grenadines and beyond. Turtle watching, hiking, nature, romance and tranquility! One of the best loved Grenada hotels!

China's princelings come of age in new leadership

BEIJING (Reuters) - In China they are known as "princelings" ? the privileged children of the revolutionary founders of the People's Republic of China. And in the generational leadership change that just took place in Beijing, it could not have been clearer that having the right family bloodlines is among the most important attributes an ambitious cadre could possess.

Of the seven men who now comprise the Communist Party's new politburo standing committee, the apex of political power in China, four are members of "the red aristocracy", led by the new general secretary of the party, Xi Jinping.

The thriving of the princelings should not be a surprise, analysts and party insiders say. Rarely in its six decades in power has the party been under more stress. Public anger over widespread corruption, widening income inequality and vast environmental degradation have chipped away at its legitimacy.

The party's over-arching goal is to maintain its grip on the nation, and moving so many princelings into top positions is akin to taking out a political insurance policy.

"Fundamentally, princelings advocate maintaining one-party dictatorship," said Zhang Lifan, a Beijing-based political commentator. "This is (their) bottom line."

The rise of the princelings comes despite the fall of one of their own ambitious brethren, Chongqing party secretary Bo Xilai, himself a one-time contender for the standing committee and a son of one of Mao Zedong's closest comrades. Earlier this year, Bo's wife was convicted of murdering a British businessman in one of modern China's biggest political scandals.

Bo himself faces possible charges of corruption and abuse of power.

But in the wrangling over the new leadership, the princelings got a boost from former president and party elder Jiang Zemin, 86, widely viewed as a backroom powerbroker. Jiang had long supported Xi's rise and helped get another princeling onto the standing committee.

Jiang sees himself as a princeling as well, party sources say. His uncle, who died in 1939, is hailed as a martyr of the revolution that brought the Communists to power in 1949. Jiang additionally hopes that backing Xi will preserve his legacy and protect his family.

Party insiders say Jiang wants to make sure his two sons, both of whom are successful businessmen, are protected at a time of enhanced scrutiny of the wealth accumulated by the families of the country's top leadership.

The new standing committee is the first to be dominated by princelings. Jiang's successor, outgoing president Hu Jintao, was the first among equals in the previous line-up, which comprised mainly technocrats and bureaucrats.

But now, according to several analysts, most senior party members have fallen in line with what late economic tsar and one-time standing committee member Chen Yun once said: "The land under heaven should one day be handed to princelings, who can be trusted not to dig the party's grave."

PRINCELING IN CHIEF

In addition to Xi, 59, those on the committee with familial ties to the country's red founders are Vice Premier Wang Qishan, who will lead the party's efforts to contain corruption; former Shanghai party secretary Yu Zhengsheng, 67, the oldest member on the committee; and Zhang Dejiang, who studied economics in North Korea and replaced Bo as party boss in Chongqing.

Beyond their commitment to party rule, insiders say the princelings' inclinations on the critical issues facing China - especially political and economic reform - are harder to discern. Xi has used standard party rhetoric since taking the top job, saying China must "continue reforming and opening up".

The princelings, analysts said, tend to be bound not by strong policy preferences, but by their privileges and the conviction they were born to rule.

"The way they rode to power is very similar, but whether they share the same outlook, the same preferences for policies, I think that's not really the case," said Damien Ma, an analyst at political risk consultancy Eurasia Group.

Some analysts are cautiously optimistic that a leadership dominated by Xi and the other princelings might move with surprising boldness.

One Beijing-based political analyst, who requested anonymity because of the sensitivity surrounding discussions on the leadership, said princelings believe it is their birthright to rule, and act accordingly. Analysts contrast them with leaders from a rival political faction, the Communist Youth League which produced President Hu.

"(The princelings) are naturally more confident and bolder than the children of commoners like Hu, whom they see as a mere caretaker, or a hired CEO," the analyst said. "The CEO is more prudent. The stakeholders are more anxious than the CEO if the company is not doing well. Princelings are likely to be bolder in pushing for change."

Some members of the political elite believe the party, after a decade of stagnation on political reform, needs to move quickly to improve government transparency, accountability and the rule of law, as well as allow more freedom of expression. They point hopefully to Xi's princeling bloodlines.

His father, Xi Zhongxun, who gave refuge to Mao during the Long March from 1934 to 1936, was a liberal. As party secretary of Guangdong in 1980, the elder Xi convinced Deng Xiaoping to allow him to set up market-oriented special economic zones in the province, the first place to do so in the Communist era. He also opposed the army crackdown on student protesters at Tiananmen Square in 1989, and championed the rights of Tibetans and other minority groups.

Others believe Xi junior's public comments and writings, however rare, indicate he and the other princelings are pragmatists.

A TALE OF TWO PRINCELINGS

Xi's ascension, along with the other members of the red aristocracy, came at an awkward moment for the princelings.

Their princeling comrade Bo Xilai was ousted in March as party boss of Chongqing, lost his seat in the wider Politburo in April and was expelled from the party in September.

But the downfall of such a high-profile princeling, analysts suggested, was not necessarily unhealthy. At a time of deepening cynicism about the leadership among many Chinese, it showed that when a princeling breaks the law, "his crime is the same as that of a lawbreaking commoner", commentator Zhang said, quoting a Chinese proverb.

The different outcomes for Xi and Bo also suggest that even for the offspring of well-connected families, the way they wield power matters. By all accounts, Xi mostly kept his head down and did what was asked of him as he rose through the party's ranks.

Bo, by contrast, was flamboyant by Chinese political standards and played the family card if he thought it could help.

Bo's father, Bo Yibo, was one of the so-called "eight immortals," and helped guide China away from some of the most disastrous policies of the Mao era. He died in 2007.

At one point before the elder Bo's death, President Hu summoned Bo and Xi and offered them the same job: to run the landlocked province of Inner Mongolia, an economic backwater.

Bo, then commerce minister, was reluctant to go and told Hu he would have to ask his father first, one party insider told Reuters. Xi, then party boss of prosperous Zhejiang province in eastern China, said he was not familiar with the ethnic issues in Inner Mongolia but was willing to go.

"It was a test, but Bo used his father to pressure Hu," the party insider said, requesting anonymity to avoid repercussions for discussing secretive elite politics. "Xi was willing to accept whatever the party arranged."

All along, Xi understood intuitively that "the higher the profile (of an up and coming official), the more difficult promotion will be", even for a princeling, said Zhang, the political commentator.

In 2000, as governor of Fujian province in the southeast, Xi gave an interview in which he quoted an ancient Chinese military strategist:

"Do not try the impossible. Do not seek the unattainable ... Do not do the irreversible. Taking up a new government post is a relay. Don't drop the baton and run your leg well."

Review: Silver Linings Playbook | Film Reviews | Gambit New ...

Ken Korman says the strength of David O. Russell's latest film is Jennifer Lawrence

Ah, the holiday season. It's the only time of year when movies with bipolar protagonists briefly seem like a perfectly good idea. Silver Linings Playbook initially presents itself as an edgy, insightful and indie-spirited film about severely damaged people desperately seeking respite for their battered souls. But it reveals its true colors in its last 20 minutes, when the movie suddenly and inexplicably transforms into the season's most conventional romantic comedy.

??Pat (Bradley Cooper) has been released from a state institution after beating his wife's lover to a pulp, and Tiffany (Jennifer Lawrence) recently got fired after sleeping with everyone in her office in the wake of her husband's death. Will they wind up in each other's arms despite their erratic behavior and the invisible obstacles that somehow keep them apart? Director David O. Russell delivered the occasional early masterwork (see 1999's Three Kings), but can't find a steady tone to pull this movie together. The oddest thing about Silver Linings Playbook is that it gets better as it goes along. That may be more a function of its initial extreme unpleasantness than any righting of a wayward ship. Lawrence (The Hunger Games) nearly saves the day with a surprisingly subtle performance. If only her character had been surrounded by a more convincing film. ? KEN KORMAN

himeshsingh: my house: Death and dying: End of Life Exit strategies

As a Humanist/Rationalist, I have always had an interest in the humanist position on end of life exit strategies.

Quite simply, this is about a making a conscious, rational decision that if diagnosed with a terminal disease or for any other reason deciding that quality of life is unacceptable, having control over the end of the life process, ensuring that the end of life happens in a quick, painless manner, rather than suffering a lingering death as the victim of a society that is dominated by relgious beliefs that prevent humans receving the same level of terminal care as a family pet.

Disclaimer: I hope to live a long and satisfying life and have no suicidal ambitions at present. Unfortunately it is the stigma of discussing end of life? that forces me to make this disclaimer as otherwise many of my friends would believe falsely that I am contemplating suicide.

I have no fear of death. Death is nothingness as the brain ceases to function. From a personal perspective, death is no different from a general anaesthetic except that you don't wake up. I remember thinking about this the last time I underwent a GA. As I was wheeled into to the prep room and the the anaesthetic was connected to the veins in my wrist, I was thinking that what I was about to experience would be identical to the moment of death in a controlled environment. That was the last thought I had before I woke up....

However, I do have a natural and understandable fear of dying. Dying is about pain and even more about loss of control as you become weaker and lose control of your own destiny. My greatest fear would be coming semi-vegatitive -suffering but unable to articulate my desire for the pain and humiliation to end.

I feel great comfort in knowing that if I am every confronted with a terminal illness that I will be able to be in control of my own destiny to the end. I am a huge support of Exit International - http://www.exitinternational.net/ a voluntary euthanasia organisation started by an Australian Doctor who actually created a patient controlled euthanasia machine more than a decade ago, but was banned from using it by the Australian Courts.

At the moment, I have two preferred strategies for end of life management. The first is self-management through use of a Helium bag (or a Helium tent for terminally ill couples). The great thing about Helium is that it doesn't produce any subjective signs of approaching hypoxia such as shortness of breath. There is no discomfort at all and unconciousness? and then death happens gradually and painlessly, within minutes as the helium replaces oxygen. However, it is important to obtain the right equipment as unfornately there are many examples of this not working when a heliumu/oxygen mix is unintentionally used instead of pure Helium. See: http://suicideproject.org/2011/08/failed-helium-exit-bag-attempt

This is wonderful blog with people trying to help each other achieve a painless death without any questioning of their intentions.

My second recommended strategy is Dignitas, the Swiss assisted suicide organisaton. http://www.dignitas.ch/index.php?option=com_content&view=article&id=20&lang=en

Dignitas provide a dying with dignity service for those with confirmed terminal illnesses. However, for most of my friends, using Dignitas would mean ending your life a long way from your family and friends, unless you could convince them to accompany you to Switzerland. I've read many wonderful stories from people arriving at Dignitas who have used their savings to pay for family and friends to accompany them to Switzlerland and have have had a wonderful final night out at a nice restaurant before ending their lives.

My strong support for a structued and and humanist end of life strategy is driven by three things. The first is my humanist worldview. Without the fantasy of religion, we can make sensible moral and ethical decisions that are in the best interests of ourselves. Secondly, I have a clear memory of the suicide of my sister, who didn't have the knowledge or support to have a painless end of life. She took rat poison and did not die painlessly. The police who recovered her body initially thought that she was murdered due to the amount of bruises on her body. The brusies were in fact caused by the reaction of the rat poison on her body.

Thirdly, I have fond recollections of my Father, who was a very caring, humanist, atheist Doctor. He had similar principles to myself in respect of end of life care for his patients. I remember one Saturday that I was was with him as he did his hospital round. He had to visit "Mr Smith", who who was an extreme diabetic with a terminal illness. All Mr Smith had wanted was was an ice-cream and was not allowed was not allowed this due to his illness. My dad bought Mr Smith an ice cream and Mr Smith passed away peacefully soon after.

I hope to die a natural death, but it is very comforting to know that if I am am ever diagnosed with a terminal illness that I will be in control of my destiny.

(AP) ? Rory McIlroy moved to the top of the leaderboard with a 5-under 67 Friday in the second round of the Dubai World Championship.

The top-ranked golfer, who missed several makeable putts on the front nine and then saw an eagle putt roll past the hole on 14, tied Luke Donald (68) and Marc Warren (67) at 11-under 133.

"I continued driving the ball well today and gave myself plenty of opportunities," said McIlroy, who has already won the European and PGA tour money titles. "Missed a couple of short ones on the front nine for birdies. But apart from that, it was a very solid round of golf and obviously in good position going into the weekend."

Sergio Garcia, playing for the first time since having laser eye surgery, matched the course record with an 8-under 64.

Garcia started with four birdies on his first five holes and then had a string of birdies at the turn before holing a long putt for eagle on 14. But he shot into the water on 16 for a triple-bogey before recovering to finish with a birdie and eagle.

"Coming back from a long break, so trying to get some good vibrations going and I managed to finish better than that," said Garcia, who is tied for 18th.

Donald joined McIlroy and Warren at 11 under with a short birdie putt on the 18th. Warren had a chance take the outright lead, but his 12-foot putt on the 18th came up short.

"Obviously, haven't dropped a shot in 36 holes, so feel good about that," said Donald, who finished with four birdies in a bogey-free round. "I had some opportunities, 10-foot, 15-footers on a few holes that I just misread. But that was a solid round of golf."

Donald said he wasn't getting his approach shots as close to the pin as the first day, but made up for it with the putter, including a difficult birdie on No. 9 and nice par save on 12.

"When I have myself a little bit of trouble out on the course, I was able to recover with the putter," Donald said.

The 189th-ranked Warren, who has not won a tournament since 2007, matched the second-ranked Donald's opening birdie and had five more, including a 12-footer on the 16th to move him into a tie with McIlroy.

"It was nice to be the Scot in the pack with the Englishman and the Northern Irishman," Warren said. "It's great to be in contention with guys like that. Looking down the leaderboard, it's a high-quality leaderboard and doesn't get any better anywhere in the world really."

It was also a good day for the South Africans as 2010 British Open champion Louis Oosthuizen (67) is only one stroke behind. He was joined at 134 by four-time tour winner Branden Grace (67).

Charl Schwartzel (67) and Richie Ramsay (68) were two shots off the pace, followed by another 10 players three strokes behind.

Several big names faltered, including fourth-ranked Lee Westwood (74). Two shots off the lead after the first round, Westwood bogeyed the 12th and 16th and then double-bogeyed the par-3 17th to fall eight shots off the lead.

Padraig Harrington, also two shots back after the first round, is five behind after a 71.

Better blood transfusions for preterm babies

ScienceDaily (Nov. 22, 2012) ? Results of new research from the University of Adelaide are a promising step forward in helping to improve the quality of life-saving blood transfusions for preterm babies, by reducing the likelihood of adverse inflammatory responses to the blood.

Blood transfusions are among the most common medical procedures experienced by preterm babies, who are often anemic and suffer blood loss.

Researchers from the University of Adelaide's Robinson Institute studied 28 preterm babies (at 28 weeks' gestation or less) who were given packed red blood cell transfusions. The results of this study are now published in the journal Pediatric Research.

"Blood transfusions are a safe and life-saving medical procedure -- they are an important part of modern-day medical care," says the lead author, Dr Michael Stark from the University of Adelaide's Robinson Institute.

"It has been suggested that blood transfusions themselves may be associated with medical complications that are unrelated to the reason for which the transfusion is given, and we don't really know why that is.

"These associations include bronchopulmonary dysplasia and necrotizing entercolitis, inflammatory conditions that affect the lungs and gut of very preterm babies."

The researchers have found a potential mechanism associated with the inflammatory response in the body.

"Within two to four hours of preterm babies receiving a blood transfusion, we have seen elevated levels of cytokines and chemokines -- signaling cells -- that stimulate inflammatory responses in the body," Dr Stark says.

"We believe that the bioactive components of packed red blood cell transfusions are initiating or amplifying these inflammatory processes in the body.

"We hope that by better understanding how the body responds to the blood, we can make improvements to blood transfusions that will reduce the likelihood of inflammatory responses. In this way, the patient will benefit from a life-saving procedure and also experience less complications as a result of that procedure.

"More research is now needed to determine exactly how this response is triggered, and how we might be able to prevent it," he says.

It takes two to tangle: Scientists further unravel telomere biology

Thursday, November 22, 2012

Chromosomes - long, linear DNA molecules ? are capped at their ends with special DNA structures called telomeres and an assortment of proteins, which together act as a protective sheath. Telomeres are maintained through the interactions between an enzyme, telomerase, and several accessory proteins. Researchers at The Wistar Institute have defined the structure of one of these critical proteins in yeast.

Understanding how telomeres keep chromosomes ? and by extension, genomes ? intact is an area of intense scientific focus in the fields of both aging and cancer. In aging, the DNA of telomeres eventually erodes faster than telomerase and its accessory proteins can maintain it, and cells die. In cancer, tumor cells hijack the process, subverting the natural method by which our bodies limit cell growth; cancer cells, then, can grow and multiply unchecked.

One of these accessory proteins, Cdc13, is integral to telomere maintenance and essential for cell viability in yeast, according to researchers at The Wistar Institute. In a study published in the journal Structure, available online now, a team of scientists led by Emmanuel Skordalakes, Ph.D., an associate professor in The Wistar Institute Cancer Center's Gene Expression and Regulation Program, has determined how mutations in a particular region of Cdc13 can lead to defects in telomeres that could jeopardize DNA.

Cdc13 normally functions as a matched-set, where two copies of the protein form what is known as a dimer. Skordalakes found that mutations in a region of Cdc13 (called OB2) prevent Cdc13 copies from binding to each other. The findings help explain the biology of this key telomere maintenance protein, and may eventually lead to novel anticancer therapeutic if their findings translate to a similar molecular system used to maintain human telomeres, Skordalakes says.

"If we could target the OB2 region of Cdc13, for example, it would throw a wrench in the works of telomere maintenance," said Skordalakes. "If you can disrupt recruitment of telomerase in humans, you could potentially drive cells to death."

Cdc13 serves a dual function in telomere replication. First, it keeps the cells' natural DNA repair mechanisms from mistaking the telomere for a broken stretch of DNA, which could cause genetic mayhem if such repair proteins fuse the ends of two chromosomes together, for example. Secondly, Cdc13 recruits telomerase and related proteins to place in order to lengthen the telomeres.

In yeast, telomeres are decorated by a multi-protein complex called CST, which contains the proteins Cdc13 (C), Stn1 (S), and Ten1 (T). Cdc13 is a key member of that complex and serves both to cap the telomere structure and recruit key enzymes.

Skordalakes' newly determined structure demonstrates that, like three of the other four regions of Cdc13, OB2 adopts what is called an oligonucleotide/oligosaccharide-binding fold (OB). These folds normally allow proteins to bind DNA or sugars, but OB2 does neither; its crystal structure indicates that this fold actually forms a large binding surface that helps two Cdc13 proteins to form a dimeric complex.

The authors then used biochemical analyses to determine that OB2 also does not directly bind the protein Stn1. Nevertheless, full-length Cdc13 OB2 mutants deficient in dimerization are also deficient in Stn1 recruitment. When the team inserted strategic Cdc13 mutations into yeast, they found that the cells had abnormally long telomeres, probably as a result of disrupted CST complex assembly caused by impaired Cdc13 dimerization.

"The dimerization of OB2 is required for the proper assembly of the CST complex at the telomeres," Skordalakes says. "When you disrupt oligomerization of this domain, you disrupt assembly of this complex, and thus regulation of telomere length."

Thanksgiving Day marked with parades, feasts in wake of Sandy

NEW YORK (Reuters) - Americans celebrated Thanksgiving by cheering parades and cooking sumptuous feasts, some sharing the bounty at East Coast emergency shelters to say thanks for what they still had after Superstorm Sandy caused widespread damage throughout the region.

"We're trying to do it Pilgrim style," said Louis DeCarolis, 51, roasting a turkey in a fire pit marked by an American flag and dug into the front yard of his son's home in Rockaway Beach, Queens, that lost power when it was flooded by the deadly storm.

Thousands of area residents are coping with the loss of homes, businesses and loved ones. At an emergency shelter in another coastal community, Belle Harbor, Queens, red cloths and red and pink roses graced tables groaning with trays of stuffing and pumpkin pies for people displaced by the historic storm that destroyed homes and businesses in New York and New Jersey.

In Texas, the crush of holiday road traffic was blamed for a massive, high-speed pileup of up to 150 vehicles on a fog-bound interstate near Beaumont that killed at least two people and injured up to 120, a sheriff's spokesman said.

New York City kicked off the holiday with cheering crowds lining streets for the 86th Macy's Thanksgiving Day Parade, the largest public event so far in a city still recovering from the storm.

Under warm and clear skies, children climbed atop police vans and on their parent's shoulders to get a better view of an enormous inflatable Hello Kitty prowling between skyscrapers along with 15 other gigantic balloons, including a 60-foot-tall Kermit the Frog balloon and a huge Charlie Brown.

The parade, which typically draws 3.5 million spectators and 50 million television viewers, also featured 28 floats, 11 marching bands, thousands of cheerleaders and dancers and Santa Claus. Celebrity performers included Whoopi Goldberg, Carly Rae Jepsen and the Muppets.

Macy's provided seats for some 5,000 people affected by Sandy, which inundated lower Manhattan with seawater, damaged shorelines and destroyed homes in New Jersey and New York. The October 29 storm killed 132 people in the United States and Canada.

Basting his turkey in the fire pit a block from the beach, DeCarolis said he planned to deliver it to a homebound neighbor and her disabled child who are still without power.

"I'd rather give it to them. We have a lot to be thankful for - we didn't lose any loved ones," said DeCarolis, a building cleaner now homeless after his basement apartment was destroyed by the historic storm surge.

New York Mayor Michael Bloomberg said the city, in partnership with local community organizations and businesses, was providing 26,500 Thanksgiving meals for people hardest hit by the storm.

About 2,000 of those meals were being served under a white tent organized by the city outside St. Frances de Sales Parish in Belle Harbor, which was dishing out another 500 meals inside the church.

"I've seen lots of smiles, warm hearts," said volunteer Carmine Iovino, 61, a retired utility worker from Queens carving a turkey alongside Long Islanders Rita Amoreno, 70, a retired nurse serving her own pumpkin cake and Ursula Lindgren, 27, a caterer, ladling her homemade soup.

Laura and William Reichard, whose nearby home is still without heat and hot water, ate their feast from a styrofoam takeout tray in the tent and said small touches like flowers on the table meant a lot after a difficult month.

"It's beautiful. It lightens up everything," said Laura Reichard, who thanked the army of volunteers. "Day and night, they've just been so wonderful."
